The aim of the study was to investigate whether the administration of warfarin before and after induction of AP in the ischemia-reperfusion model improves the clinical course of this disease. The studies were conducted on male Wistar rats. Acute hemorrhagic necrotizing pancreatitis was induced by pancreatic hypoxia for 30 minutes followed by reperfusion. The research was carried out in 2 stages. The first step was to determine the effect of warfarin in causing AP. In the second step, warfarin was administered after the induction of acute pancreatitis. The following markers were assessed: histological evaluation of pancreatic specimen, concentration of d-dimer, IL-1β, plasma amylase and lipase activity, and pancreatic blood flow. Low and medium doses of warfarin administered 7 days before the induction of acute pancreatitis have a positive effect on the course of inflammation. However, excessive doses exacerbate inflammation and deepen the studied parameters. Administration of medium and high doses of warfarin after the induction of AP reduces plasma activity of pancreatic enzymes, levels of d-dimer and IL-1β. It also reduces the intensity of morphological and inflammatory changes in the pancreas. It improves pancreatic blood flow and accelerates pancreatic healing. The results demonstrate the usefulness of warfarin in the prevention and treatment of acute pancreatitis.
