In this work, the Metal-Organic Frameworks (MOFs) called Fe-MIL-101-NH2 containing iron ions and aminoterephthalic acid in their structure were employed. Isoniazid (INH) antituberculosis drug was loaded into MOF porous structure. This INH-MOF complex was embedded into a inhalatory blend of PLGA and leucine microparticles prepared by spray drying. A factorial experimental design study 23 was used to optimize the formulation. Powder aerodynamic properties were assessed using a twin stage impinger based on the emitted dose (% ED) and the fine particle fraction (% FPF). The INH content was 3.7% w/w. Magnetic resonance imaging (MRI) contrast capabilities were tested on porous lung tissue phantom and ex vivo rat lungs In vitro cell uptake and oxidative stress studies were performed on macrophage (RAW) cell lines. The final blend showed good aerodynamic properties (% FPF = 56.7%), modified drug release, ability to penetrate into macrophages (INH uptake after 24 h is 21% of the dose) and promising contrast properties (positive contrast) enabling monitoring of the formulation distribution in heterogeneous lung structure. The paper presents a complete research, starting with the raw MOF material, developed a fully functional teranostic inhalatory formulation with potential use in personalized therapy of pulmonary tuberculosis.
